Polysaccharide-based chiral stationary phases (CSPs) are widely used in the pharmaceutical, agricultural, and natural product industries. The core of polysaccharide CSPs consists of a silica gel support material, a polysaccharide backbone, and a chiral selector. The support material plays an important role in providing CSPs with rigidity and robustness that can be used in high-pressure applications. The chiral selector and the polysaccharide backbone are responsible for distinguishing between enantiomers through a series of intermolecular interactions. These interactions including hydrogen bonding, π-π stacking, dipole forces, inclusion, and exclusion can be exploited to separate enantiomers. In a chiral column, the potential combination of interactions is able to separate enantiomers. The type and frequency of these interactions are highly dependent on the type of polysaccharide backbone, the functionalization of the chiral selector, and the 3D structure formed by supporting on the silica gel. In addition, the solvation, swelling, or shrinking of the derivatized polymer backbone in the presence of certain solvents or additives plays an important role.
Polysaccharide derivatives can be physically coated onto a chromatographic support, allowing a large surface area and high efficiency. The coated polysaccharide CSPs can operate in normal phase, polar organic, and reverse phase elution modes. However, due to this manufacturing technique, the range of solvents used is limited. Some strong solvents, such as tetrahydrofuran, dichloromethane, and chloroform, although contributing to the solubility of the compound, may destroy the coated CSPs. Therefore, attention should be paid to the choice of mobile phase before use.
Amerigo Scientific provides normal phase HPLC columns whose stationary phase is spherical silica gel coated with the polysaccharide derivative. Columns of various size are available, from semi-micro to preparative columns. We also provide chiral HPLC columns packed with 3μm CSPs. Compared with the 5μm CSPs- and 10μm CSPs-based columns, the columns with 3μm CSPs show higher column efficiency and better separation. More than 80% of enantiomers can be separated with our polysaccharide-based chiral columns.

Amerigo Scientific provides reversed phase HPLC columns that are packed with the same chiral polysaccharide derivatives as the normal-phase HPLC column, but the used silica gel is different. Our reversed phase HPLC columns are suitable for the separation of water-soluble samples, aqueous preparations, or samples that need to be detected using mass spectrometry (MS). We also provide columns with an inner diameter of 2.1mm for LC/MS.
