Stem cell therapy is one of the fastest growing fields of research, which has the potential to treat many severe diseases due to the unique properties of stem cells with self-renewal and differentiation potential. Transfection can be applied to reprogram somatic cells and to engineer stem cells. Biological states of stem cells such as differentiation, self-renewal, and growth can be controlled by introducing double-stranded DNA and double-stranded or single-stranded RNA. In addition to facilitating gene expression, transfection and transduction can be used to downregulate gene expression levels in stem cell to regulate functions of the cells.
Gene transfection methods are generally classified into viral and non-viral. Viral vector-based transfection methods are widely used because of their high efficiency and long-term maintenance of expression, although their limitations include immunogenicity, carcinogenicity, limiting DNA load, and high cost. An ideal transfection method should prevent degradation of the delivered gene, penetrate the target-cell membrane, and allow the gene to insert into the nucleus without causing damage to the cells. As one of the basic materials for nonviral vector transfection, liposomes meet several criteria for the ideal transfection agent. Various natural and synthetic liposomes can be used to facilitate DNA or RNA encapsulation.
Amerigo Scientific provides lipid-based transfection reagents for stem cells. These reagents are composed of synthetic cationic lipids that are usually mixed with helper lipids. At physiological pH, the lipid mixture assembles into liposomes or micelles with positive charge, and can form complexes with negatively charged nucleic acid via electrostatic interactions. Then these complexes are mainly internalized by endocytosis.
