  • Enzymes are biocatalysts that are used in various fields, due to their activity, selectivity and specificity. However, the structure of enzymes is unstable and their activity is easily affected by conditions. In order to allow for the wide industrial application, enzymes are immobilized on various carriers. The immobilized enzymes are attached to specific solid supports and are reusable over successive catalytic cycles. Compared to the native enzymes, the immobilized enzymes show increased activity in organic solvents, are more stable under various reaction conditions, and their use can simplify process. In addition, the immobilized enzymes are easily separated from products, resulting in products of higher purity. The reaction can be easily controlled by removing the immobilized enzymes from the reaction mixture. Due to their operational and economic advantages, immobilized enzymes are used in the production of active pharmaceutical ingredients (APIs), food ingredient preparation, chemical compound synthesis, and many other applications.

    In enzyme immobilization technology, the active group, microenvironment, shape of the carrier and other factors may affect the affinity of the carrier and the enzyme, the activity, stability, reusability and recyclability of the immobilized enzyme. Generally, carrier materials have the characteristics of a large specific surface area, a certain pore diameter, good hardness, mechanical strength, and easy surface modification or coating. The selection of the carrier is very important for the effective immobilization of the enzyme.

    Epoxy Resins

    Seplife® epoxy resins are porous, robust, and hydrophilic, and allow simple and rapid enzyme immobilization. The epoxy acrylate resins are designed to form very stable covalent bonds with different protein groups (amino, mercaptan, phenol) under mild pH and temperature conditions.

    Amino Resins

    Seplife® amino acrylate resins are porous, robust and hydrophilic. These amino resins are functionalized with primary amines, and the covalent enzyme immobilization is performed by pre-activation with glutaraldehyde. The aldehyde group reacts with the amino group of the enzyme to form a Schiff base, which is very stable in the pH range of 3-8. The amino acrylate resins are designed to form very stable covalent bonds with different protein groups (amino, mercaptan, phenol) under mild pH and temperature conditions.

    Affinity, Ionic and Adsorption Immobilization Resins

    Amerigo Scientific also offers resins for adsorption immobilization, ionic immobilization, or affinity immobilization. Adsorption immobilization resins are suitable for enzymes that need to be used in oil and organic solvents, and affinity immobilization resins are suitable for recombinant enzymes containing His-Tag. Enzyme ion immobilization is a method of immobilizing enzymes by ion interaction, which depends on the isoelectric point of the enzyme, the optimal activity pH of the enzyme, and the ionic strength of the immobilization buffer.
