Affinity chromatography is a protein separation method based on specific interactions between biological molecules (such as enzymes and substrates, receptors and ligands, antibodies and antigens). The specificity and reversibility of these interactions can be utilized to achieve protein purification.
Amerigo Scientific offers a variety of high-quality and reliable affinity resins for the purification of His-tagged proteins, antibodies, proteins containing Fc, or mRNA.
Amerigo Scientific offers two product series suitable for metal affinity purification of His-tagged proteins. These resin products are prepared by functionalizing Seplife® 6AG beads containing 6% cross-linked agarose with iminodiacetic acid (IDA) or nitrilotriacetic acid (NTA) through stable covalent bonds. The resins are supplied in the form of sodium ions (Na+) or nickel ions (Ni2+), or can be modified with other metal ions as required. The metal capacity of the resins is varying as follows: copper ions (Cu2+) > nickel ions (Ni2+) > zinc ions (Zn2+) > cobalt ions (Co2+).
Ni2+ affinity resins are one of the most widely used metal chelate chromatography resins for purifying proteins with His-tags. According to the chelating agent used to immobilize the Ni2+, Seplife® 6AG Ni2+ affinity resins can be divided into two types: Ni-lDA and Ni-NTA. A key characteristic of Ni-IDA resins is their higher binding capacity compared to Ni-NTA resins. The stronger chelation of the Ni2+ by NTA results in a stronger binding ability of the Ni-NTA resin and higher stability against high concentrations of reducing agents. This is a critical advantage for applications requiring high purity and the absence of contaminating metal ions.

rProtein A Seplife® Suno resin can specifically bind to the Fc region of antibodies as well as recombinant proteins containing Fc region. Therefore, it can be used for the separation and purification of these antibodies or proteins. Using rProtein A Seplife® Suno resin, it is possible to obtain highly pure biomolecules from complex samples (such as serum or fermentation broth) in a single capture step.

Seplife® Oligo dT(20) affinity resin is a 50-micron polymeric resin with very uniform particle sizes, and is designed for the purification and isolation of mRNA produced via in vitro transcription (IVT). The resin is based on crosslinked polystyrene divinylbenzene functionalized with oligo dT20. With high selectivity and capacity, Seplife® Oligo dT(20) affinity resin can be used to efficiently separate mRNA from enzymes, plasmid DNA, and other IVT reaction components. This resin is ideal for large-scale mRNA purification, supporting vaccine and gene therapy development.
