Membrane proteins are large biological molecules that are embedded in or attached to the membrane of a cell or an organelle inside the cell. Membrane proteins are key players in biological processes, responsible for the transport of molecules into and out of cells, the maintenance of the electrical potential difference across the membrane necessary for ion conduction and active transport, the sensing and transduction of intracellular and extracellular signals, the coordination of intracellular signaling, and cell adhesion.

Mutations in membrane proteins have a wide range of effects, some of which are related to the properties of the proteins. Missense mutations (also called nonsynonymous mutations) may disrupt the native structure of a protein by causing steric clashes or affecting primary, secondary and tertiary structures of the protein. In addition, missense mutations may disrupt protein-protein or protein-membrane interactions. The missense mutations in signal peptides may affect protein trafficking and embedding into the membrane. Many diseases are caused by mutations in membrane proteins, and in particular mutations that disrupt the folding or assembly of membrane proteins are thought to be a major cause of human diseases.
