  • Human platelet lysate (HPL) is a multifactorial supplement obtained from human platelets, which contains abundant growth factors and cytokines necessary for cell growth and proliferation. In a wide range of applications, HPL is considered a good substitute for animal serum. The replacement of fetal bovine serum (FBS) with HPL can reduce the immunogenicity and remove the transmission of zoonotic diseases, which is beneficial to the clinical application of cell proliferation or production.

Two main differences in HPL protein content compared to FBS are the higher content of immunoglobulins and the possible presence of fibrinogen and other coagulation factors, when HPL is produced without thrombin activation.

Mean Protein Content in HPL and FBS
Total Protein Albumin Immunoglobulin G Immunoglobulin A Immunoglobulin M Fibrinogen
    HPL* 55-65 mg/mL 35-40 mg/L 8-12 mg/mL 1-2 mg/mL 0.5-1.5 mg/ mL 1.0-3 mg/mL**
    FBS 2-70 mg/mL 20-36 mg/mL  -  -  -  -

    * HPL prepared from a platelet concentrate obtained stabilized in plasma; ** HPL obtained by freeze-thaw or S/D processes

HPL contains high concentrations of growth factors, such as insulin-like growth factor-1 (IGF1), platelet derived growth factor (PDGF)-AA, -BB, -AB, transforming growth factor (TGF)-β1, brain derived neurotrophic factor (BDNF), epidermal growth factor (EGF), vascular endothelial growth factor (VEGF), hepatocyte growth factor (HGF), and basic fibroblast growth factor (bFGF). The contents of these growth factors range from 0.1 to 300 ng/ml, and TGF-β1, PDGF-AB, IGF1, and BDNF are generally higher in content than other growth factors.

HPL, like FBS, contains minerals such as sodium, potassium, magnesium, iron, as well as glucose and vitamins that can influence cell growth, differentiation, and function.

    Product Range

Amerigo Scientific offers research-grade and GMP-grade HPL products derived from platelets of healthy voluntary donors at certified blood banks. All HPL products are naturally free form other animal derived material. No animal serum is added during the manufacture of these products. Two formats are available according to the need of applications, as follows:

    1. A standard HPL that requires the addition of heparin before use.
    2. A fibrinogen-depleted HPL that does not require the addition of heparin prior to use.

    • Research Grade HPL

HPL ELAREM™ Ultimate FDi, Gamma Irradiated, fibrinogen depleted, no heparin required a virus-inactivated and fibrinogen-depleted HPL of US origin cell culture

    Key Benefits

    • A suitable alternative to FBS without the risk of xenogeneic immune reactions and transmission of bovine prion
    • High performance - Use at lower concentrations, such as 2.5%, 5%, or 10%
    • Low batch-to-batch variation - Standardized product from multiple donors
    • Wide applicability - Used for culturing many types of human cells, such as leukemia cell lines, dermal fibroblasts, Hela cervical cancer cells

    Quality Assurance

    • All batches are fully tested for pathogens according to FDA guidelines and found non-reactive for HBsAg, HIV I/II antibody, CMV, HCV- and HBC-antibody, HIV-, HCV-, HBV-, HAV-, PVB19-NAT, Syphilis and Treponema pallidum.
    • Each batch is filtered to 0.1 micron before being dispensed, aseptically, into either bottles or cryo-bags.
    • Each batch of HPL is tested for the absence of bacteria, fungi, yeast, and mycoplasma.
    • Biological performance of final batches is assessed by cell culture on mesenchymal stem cells, fibroblasts, and primary cells.
