In the in vitro mammalian gene mutation test, commonly used cell lines include L5178Y mouse lymphoma cell, Chinese hamster lung (V79), Chinese hamster ovary (CHO) cells, and TK6 human lymphoblastoid cells. The in vitro mammalian gene mutation test can be used for the detection of gene mutations in thymidine kinase (TK), hypoxanthine-guanine phosphoribosyltransferase (HPRT), or xanthineguanine phosphoribosyltransferase (XPRT). Mutations of HPRT, TK and XPRT occur in different spectra of genetic events.

Comet assay, also known as single-cell gel electrophoresis assay, is a method to determine the genotoxicity of a substance by detecting DNA strand damage. Amerigo Scientific offers easy-to-use and reliable comet assay kit that can be used to effectively detect and quantitatively analyze DNA single‐strand breaks, DNA double‐strand breaks, damage to DNA bases, DNA interstrand cross‐links, and apoptotic fragments caused by genotoxic substances.
In the comet assay, single cells are embedded on microscope slides with agarose gels. After electrophoresis, DNA is stained using fluorescent dyes and visualized using a fluorescence microscope. DNA fragments constitute the comet tail, and the length and fluorescence intensity of comet tail can be analyzed to determine the degree of DNA damage.
