  • Genotoxicity testing in mammalian cells is important for assessing the potential carcinogenic risk of substances and is a crucial step in drug development, chemical safety assessment, and environmental risk assessment. Compared to bacteria or other simpler systems, mammalian cells are genetically and physiologically more similar to human cells, and this similarity increases the predictive value of tests for human effects. Conducting in vitro mammalian cell genotoxicity tests aims to identify a series of genetic alterations caused by tested substances, including gene mutations, structural chromosomal aberrations, numerical chromosomal aberrations, and DNA breaks.

Amerigo Scientific offers a range of high-quality assay kits designed to overcome the challenges of in vitro mammalian cell-based genotoxicity testing by simplifying the assay process, eliminating tedious preparation, and enhancing the reliability through refined product specifications.

    Product Characteristics

    • Convenience: The use of our kits reduces the time for preparing induced S9 and reagents, thereby greatly shortening the experimental cycle.
    • Accuracy: Each component in the kits undergoes strict quality testing to ensure that the test results are accurate, reliable, and reproducible.
    • Stability: Our test kits are highly stable and are easy to transport and store.
    • Wide application: Our test kits can be used for genotoxicity testing of food, drug candidates, cosmetics, chemicals, pesticides, and other substances.

In Vitro Mammalian Cell Gene Mutation Test Kits

In the in vitro mammalian gene mutation test, commonly used cell lines include L5178Y mouse lymphoma cell, Chinese hamster lung (V79), Chinese hamster ovary (CHO) cells, and TK6 human lymphoblastoid cells. The in vitro mammalian gene mutation test can be used for the detection of gene mutations in thymidine kinase (TK), hypoxanthine-guanine phosphoribosyltransferase (HPRT), or xanthineguanine phosphoribosyltransferase (XPRT). Mutations of HPRT, TK and XPRT occur in different spectra of genetic events.

    TK Gene Mutation Test Kits

The TK gene mutation test is a genotoxicity test to detect autosomal gene mutations in mammalian cells cultured in vitro caused by test substances. In our TK gene mutation test kit, L5178Y TK+/- mouse lymphoma cells are as the test system. The kit contains the main reagents and cells for the TK gene mutation test.

    HGPRT Gene Mutation Test Kits

The HGPRT gene mutation assay is an in vitro method for the detection of gene mutations on the X chromosomes of mammalian cells induced by test substances. Our HGPRT gene mutation test kit uses V79 cells as the test system and provides the main reagents and cells for the test.

In Vitro Mammalian Cell Chromosome Aberration Test Kits

The in vitro mammalian cell chromosome aberration test is an in vitro genotoxicity test used to assess the ability of substances to induce structural chromosome aberrations in cultured mammalian cells. Our in vitro mammalian cell chromosome aberration test kit uses Chinese hamster lung (CHL) cells as the detection system and contains the main reagents and cells.

In Vitro Mammalian Cell Micronucleus Test Kits

The in vitro mammalian cell micronucleus test is a genotoxicity test to determine whether a substance is genotoxic to cells in culture by the detection of micronuclei (MN) in the cytoplasm of interphase cells. Our in vitro mammalian cell micronucleus test kit uses Chinese hamster lung (CHL) cells as the test system, and the main reagents and cells for the micronucleus test in vitro are included in the kit.

    Comet Assay Kits

Comet assay, also known as single-cell gel electrophoresis assay, is a method to determine the genotoxicity of a substance by detecting DNA strand damage. Amerigo Scientific offers easy-to-use and reliable comet assay kit that can be used to effectively detect and quantitatively analyze DNA single‐strand breaks, DNA double‐strand breaks, damage to DNA bases, DNA interstrand cross‐links, and apoptotic fragments caused by genotoxic substances.

In the comet assay, single cells are embedded on microscope slides with agarose gels. After electrophoresis, DNA is stained using fluorescent dyes and visualized using a fluorescence microscope. DNA fragments constitute the comet tail, and the length and fluorescence intensity of comet tail can be analyzed to determine the degree of DNA damage.
