RNA synthesis kits are convenient laboratory tools used to synthesize RNA in vitro. In the presence of ribonucleoside triphosphate (rNTP), the template DNA is transcribed by T7, T3, or SP6 RNA phage polymerase. The polymerase moves along the DNA template strand in the 3' to 5' direction and uses base pairing complementarity with the DNA to synthesize an RNA strand in the 5’ to 3’ direction. This is followed by modification of the synthesized RNA. The mRNA synthesized in eukaryotic organisms is co-transcriptionally modified by the addition of the AA cap structure, which protects the transcript from nuclease digestion and facilitates translation. Alternatively, cap structure analogs can be used to co-transcriptionally cap RNA transcripts during synthesis. After successful in vitro transcription, the RNA products need to be obtained by degradation of the template and affinity purification of the synthesized RNA. Synthesized RNA can be used for a variety of applications including RNA structure and function studies, nuclease biochemistry, RNase protection assay probes and hybridization-based blotting, antisense RNA and RNAi experiments, microarray analysis, microinjection, in vitro translation, and vaccine development.
