Custom Probes for qPCR

Probes used in real-time PCR (quantitative PCR, qPCR) are synthetic oligonucleotides and can be manufactured using natural DNA bases or RNA bases, and can comprise artificially modified bases, or a variety of pendant chemical groups. Although most needs are well met using unmodified oligodeoxynucleotides, the use of chemical modifications can improve the qPCR performance in many areas. In qPCR, probes can be used to quantify target sequences accurately and rapidly due to their ability to bind specifically to the target.

Amerigo Scientific offers a wide range of probes suitable for qPCR and reverse transcription quantitative PCR (RT-qPCR) experiments. A variety of fluorophores and quenchers are available, such as VIC, NED, and PET dyes. Customized probes can be shipped within 5 to 7 working days.

Scale, Yield and Method

We provide qPCR probe oligonucleotide at three synthetic scales: 40 nmol, 200 nmol and 1000 nmol. Prices include synthesis of maximum 40 nucleotides.

40 nmol scale: approx. 5 nmol min. yield
200 nmol scale: approx. 20 nmol min. yield
1000 nmol scale: approx. 60 nmol min. yield

All qPCR probes are purified by HPLC.

Format and Concentration

Probes can be delivered in any form, concentration, or combination. Dried, dissolved, or mixed forms are available. Concentrations can be customized upon request.

In the standard form, probes are dissolved in a tube at a concentration of 100 pmol/μl. Available buffers include H2O buffer and Tris buffer.


All probes are delivered with a specification sheet containing information on GC-content, molecular weight, melting temperature (Tm), OD value, sequence, and quantity (μg and nmol).


A rigorous quality control system ensures the highest quality standards of synthetic oligonucleotides. Before synthesis, all chemicals are tested to ensure that they meet quality standards. In fully automated oligonucleotide synthesis, all steps of synthesis are monitored by multiple control functions on the synthesizer, ensuring that the coupling efficiency of the synthesis meets the requirements.

The synthesis is followed by further quality control to ensure the quality of the oligonucleotides. All oligonucleotides are routinely analyzed by optical density (OD) measurements. In addition, the oligonucleotides are randomly analyzed by gel electrophoresis and liquid chromatography-mass spectrometry (LC-MS). All the steps are performed to ensure that the quality of the synthesized oligonucleotides meets the requirements.
