Although most needs are well met using unmodified oligodeoxynucleotides, the use of chemical modifications can improve the qPCR performance in many areas. In qPCR, probes can be used to quantify target sequences accurately and rapidly due to their ability to bind specifically to the target.

The synthesis is followed by further quality control to ensure the quality of the oligonucleotides. All oligonucleotides are routinely analyzed by optical density (OD) measurements. In addition, the oligonucleotides are randomly analyzed by gel electrophoresis and liquid chromatography-mass spectrometry (LC-MS). All the steps are performed to ensure that the quality of the synthesized oligonucleotides meets the requirements.
