In Vitro Mammalian Cell Micronucleus Test (without CHL)utilizes Chinese hamster lung (CHL) cells as the test system for detecting the mutagenic potential of chemical substances. Under conditions with and without metabolic activation systems, CHL cells are exposed to the test substance. Subsequently, the cells are treated with Cytochalasin B (cytoB), an inhibitor of actin polymerization, to achieve the optimal frequency of binucleated cells. The cells are then harvested, prepared on slides, stained, and analyzed under a microscope to determine the micronucleus rate in cells that have completed one mitotic division (binucleated cells).
