Gene knockout (KO) cells are genetically engineered cells in which a specific gene has been permanently inactivated or deleted, preventing the expression of its corresponding protein. They are one of the key tools in functional genomics, used to study disease mechanisms, drug responses, and the biological functions of specific genes.
A wide range of cells can be engineered utilizing gene-editing technologies to ensure precise disruption of the target gene. The most common methods employed include the CRISPR-Cas9 system, transcription activator-like effector nucleases (TALENs), and zinc finger nucleases (ZFNs). The CRISPR-Cas9 system is currently the gold standard in genome editing. By utilizing a specifically designed guide RNA (gRNA) to direct the Cas9 endonuclease to the target locus, double-strand breaks (DSBs) are introduced. The cell's natural error-prone repair mechanism creates insertions/deletions, leading to a premature stop codon and functional knockout. TALENs are engineered restriction enzymes that offer high specificity and precision, exceptionally useful for editing complex genomic regions while strictly minimizing off-target effects. ZFNs utilize engineered DNA-binding domains fused to a cleavage domain to create targeted DSBs to successfully disrupt the target gene.
KO cell products are broadly classified into cell pools and cell lines based on their population types. KO cell pools are heterogeneous populations of cells harvested shortly after the gene-editing process, and contain a mix of wild-type, monoallelic, and biallelic knockout cells. They have the advantages of faster generation and high cost-effectiveness, making them ideal for initial rapid screening, preliminary target validation, or situations where strict requirements for complete protein absence are not necessary. KO cell lines are pure populations derived from a single edited cell. They provide a 100% pure genetic background with complete ablation of the target protein, so they are the gold standard for rigorous mechanistic studies and definitive antibody validation.
