The CacoReady plate can provide an in vitro cell model based on 21-day differentiated Caco-2 cells, which resemble the intestinal epithelial barrier. The transwell version of CacoReady can be used to assess intestinal permeability. In a non-polarized form, it is used for toxicity assessment.
Caco-2 is an immortalized cell line that originates from human colon and rectal cancer cells and can differentiate and have a structure and function very similar to that of small intestinal epithelial cells when cultured in vitro, enabling the simulation of the absorption and transport process of small intestinal epithelial cells. Caco-2 cell-based in vitro model are widely used in new drug development and research on the mechanism of drug intestinal absorption.
Using CacoReady, Caco-2 cells are cultured on a transwell plate with a permeable membrane support for 21 days, forming apical and basal compartments, which simulate the intestinal lumen and bloodstream. Caco-2 cells also express key efflux transport proteins such as multidrug resistance protein 1 (MDR1) and breast cancer resistance protein (BCRP), making CacoReady a tool for studying active drug transport.

HT-29 cells are a secretory human colorectal adenocarcinoma cell line that can produce mucus upon induction, focusing on simulating the intestinal barrier and mucosal environment. Compared to the standard Caco-2 culture, CacoGoblet combines the absorptive (Caco-2) and mucous-secreting type (HT29-MTX) cells, creating a model with physiological intestinal characteristics. This co-culture forms a protective mucus layer lining on top of the cells, forming a less tight barrier than pure Caco-2 culture and exhibiting more physiological electrical properties similar to human small intestine.
In the CacoGoblet plate, the cell model after 21 days of cultivation of Caco-2 and HT-29 cells has a phenotype of absorption and mucous secretion that makes it extremely similar to the physiological condition of intestinal epithelium. Due to its physiological relevance, CacoGoblet can provide valuable insights into passive diffusion and active transport, often revealing higher permeability, especially for compounds with low permeability. In addition, CacoGoblet responds well to inflammatory mediators and can be used to evaluate the efficacy of anti-inflammatory drugs.

MDCKII cells originate from the distal tubular epithelial cells of canine kidneys, characterized by high transepithelial electrical resistance and strong transport capabilities. Using PreadyPort, after inoculating MDCKII cells and culturing them on a transwell plate with a permeable membrane support for 11 days, a polarized morphology with distinct brush border structure, apical cell-cell junctions, and lateral space will be formed. Intercellular tight junctions will also be formed.
MDCKII cells support transient and stable gene transfection, making them particularly suitable for predicting the in vivo drug brain penetration and transport-mediated drug-drug interactions. Their versatility has enabled its wide application in permeability studies. Additionally, this model is also used to characterize transcellular transport mechanisms and the effects of tight junction proteins, osmotic changes, and ion/size selectivity.

PreadyTake provides a ready-to-use solution for testing drug-transporter interactions with HEK293 cells expressing uptake carriers. A 96-well plate is used for 11-day cultivation of HEK293 cells that overexpress MATE1, OCT2 or OATP1B3.
