    In the chemical synthesis of oligonucleotides, one of the key steps is the construction of an internucleotide linkage. To date, many synthesis technologies have been developed for the formation of internucleotide bonds, such as phosphodiester, phosphotriester, phosphite, phosphoramidite, and H-phosphonate methods. Among these methods, the phosphoramidite method is the most widely used.

    The phosphoramidite method generally enables higher speed and higher yield in internucleotide bond formation compared with the other methods. Phosphoramidites used as building blocks in the phosphoramidite method are easily prepared and stable to store. Therefore, the phosphoramidite method is often used in the synthesis of oligonucleotides, and enables the production of a variety of biologically important substances, such as primers for the polymerase chain reaction, oligonucleotide probes, oligonucleotide medicines, and cyclic dinucleotides.

    Amerigo Scientific offers natural and modified nitrogenous base and high-efficiency phosphoramidite chemistry reagents, such as acetylating reagents, activation reagents, phosphorylating reagents, etc. In addition, N-Hydroxysuccinimide (NHS) esters are available for the functionalization of an amino moiety in oligonucleotides.
