Affinity chromatography is one of the most diverse and powerful chromatographic methods for purifying a specific molecule or group of molecules from a complex mixture. It is based on highly specific biological interactions between two molecules, such as between an enzyme and a substrate, a receptor and a ligand, or an antibody and an antigen. These interactions are usually reversible and are purified by placing one of the interacting molecules, called affinity ligands, on a solid substrate to form a stationary phase while the target molecule is in the mobile phase.
