Introduction to Isotype Controls

Isotype controls are used to verify the specificity of the primary antibody by eliminating background staining resulting from non-specific binding of the antibody to the cell surface. Typically, isotype controls require the same species, subtype, dose, and immunoglobulin of the same origin and subtype as the primary antibody. Moreover, it can be used as a competitive binding antibody that performs the same function as a functional blocking antibody. Main Applications

Flow cytometry

We generally recommend that customers need to use isotype controls for the following situations:

• Customers who are not very familiar with flow cytometry.
• Customers who are interested in intracellular flow cytometry analysis, such as intracellular cytokines, chemokines, and signaling proteins analysis.
• Customers using uncommon markers or less specific markers. In this case, the location of positive cells can be determined and false positives can be ruled out by suitable isotype controls.

Immunohistochemistry

Appropriate isotype controls are essential for accurate interpretation of Immunohistochemistry results. In our labs, we advise customers to set up isotype controls to support the validity of the stain and to ensure that the observed staining patterns are true, accurate, and reliable.

Immunofluorescence

For direct staining that labels primary antibodies, we recommend replacing primary antibodies with isotype controls that label the same fluorophores to help ensure that all detection signals are derived from the specific binding of primary antibodies to antigens. The indication of antibody binding to the target protein in the sample is only valid when the chromogenic signal of the isotype control group is weaker than that of the specific antibody group. Furthermore, isotype controls can also help determine whether the second antibody has cross-reactivity.
