BACKGROUND: His-tag, short for histidine tag, is a short sequence of histidine residues (typically 6 to 10 histidines) added to proteins to facilitate their purification and detection. It is widely used in molecular biology and biochemistry due to its simplicity and effectiveness. The His-tag consists of a sequence of histidine residues, often written as (His)₆ when six histidines are used. This sequence is usually added to either the N-terminus or C-terminus of a protein of interest during the cloning and expression process.
