Background

What Are Epitope Tags

Epitope tags are short amino acid sequences that are commonly fused to a protein of interest to facilitate the detection, purification, and localization of the tagged protein. These tags are often added to the N- or C-terminus of a protein using recombinant DNA technology, allowing researchers to easily study the tagged protein in various experimental settings. Epitope tags are particularly valuable for studying proteins that are difficult to detect or purify. Some commonly used epitope tags include the hemagglutinin (HA) tag, Myc tag, FLAG tag, and green fluorescent protein (GFP) tag. Tag antibodies can specifically recognize and bind to these epitope tags, allowing for the detection and analysis of the tagged protein.

Common Tags Overview

  • Flag-tag: 8 amino acid peptide, DYKDDDDK.
  • HA-tag: A surface glycoprotein of HA protein consists of amino acids 98-106, YPYDVPDYA.
  • His-tag: Polyhistidine consists of at least six histidine (His) residues.
  • Myc-tag: A polypeptide (EQKLISEEDL) derived from the c-myc geneV5-tag.
  • Stre-tag: A peptide has a binding affinity towards streptavidin.
  • V5-tag: A peptide sequence (GKPIPNPLLGLDST) derived from simian parainfluenza virus type 5.
  • GFP: A fluorescent protein tag.
