What Are Immunohistochemistry Antibodies?

Types of Immunohistochemistry Antibodies

Primary Antibodies

Bind directly to the target antigen in tissues. Usually monoclonal or polyclonal antibodies.

  • Monoclonal antibodies: A single clone derived from B cells that is highly specific for a single epitope on an antigen.
  • Polyclonal antibodies: Antibodies produced from different B cell clones, recognizing multiple epitopes on the same antigen with higher sensitivity but less specificity.

Secondary Antibodies

  • Conjugate to primary antibodies. It is often conjugated to detectable markers, such as enzymes (e.g. horseradish peroxidase or alkaline phosphatase) or fluorescent dyes (e.g. FITC or APC).
  • Amplify the signals and enhance the visibility of antigen-antibody complexes.

Ensure that the antibodies recognize specific and unique epitopes on the target antigen. Monoclonal vs. Polyclonal:
Monoclonal antibodies are preferred due to their high specificity.
Consider the host species in which the antibody is produced. The selection of the secondary antibody needs to be specific to the host species of the primary antibody. Ensure the antibody has been validated for use in the specific tissue type and species you are studying.
