Background

Introduction of Activating Antibodies

A desired and defining feature of an activating antibody is its ability to bind and activate the target receptor, thereby mimicking the activity of the native ligand. The propensity of an antibody to function as an activator can be influenced by several factors, including the binding epitope, affinity, valency, degree of receptor occupancy, and the interaction of the antibody crystallizable fragment (Fc) domain with Fcγ receptors (FcγRs). Unlike antagonists, which generally aim to achieve the highest possible binding affinity to competitively inhibit the ligand-binding domain of the receptor, there are no definitive rules that can predict an antibody's potential to act as an activator. Consequently, the identification and selection of activating antibodies have largely been driven by empirical functional characterization using cell-based and in vivo model systems. However, a detailed understanding of the specific stoichiometric and binding properties of the native ligand-receptor complex can aid in designing an antibody with optimal activating function against a specific receptor. Although receptor structures, ligand-binding, and signaling-engagement mechanisms are generally conserved within receptor families, they are quite unique between families.

For example, the T cell-specific receptor CD28 plays a crucial role in regulating the immune system. The engagement of CD28 with its ligands, CD80/CD86, on antigen-presenting cells, delivers a crucial costimulatory signal alongside the T cell receptor for effective T cell activation. This costimulatory signal can also be mediated by the engagement of CD28 with its activating antibody.
