Fc silencing typically involves genetic engineering techniques and strategies. For example, exchanging or swapping certain amino acids between two Fc molecules derived from different IgG subclasses is a strategy that can be employed to silence antibody effector functions. This can disrupt the binding to Fc receptors, effectively silencing the effector functions of the antibodies. Another strategy for silencing antibody effector functions is through the introduction of point mutations. By mutating one or several of these amino acids, the interaction can be disrupted, leading to a reduction or complete loss of the effector functions. The Leu234Ala/Leu235Ala (LALA) mutations are common mutations used for Fc silencing. In addition, modifying the glycosylation pattern in the Fc domain can also impact the effector functions of the antibody.
