Why Silence Fc Domains?

Fc Silencing Strategies at Amerigo Scientific

Another strategy for silencing antibody effector functions is through the introduction of point mutations. By mutating one or several of these amino acids, the interaction can be disrupted, leading to a reduction or complete loss of the effector functions. The Leu234Ala/Leu235Ala (LALA) mutations are common mutations used for Fc silencing. In addition, modifying the glycosylation pattern in the Fc domain can also impact the effector functions of the antibody.
