Biomarkers are biological molecules in blood, other body fluids, or tissues that can be measured and evaluated as indicators of normal biological processes and pathogenic occurrence and progression. They can also be used to assess pharmacologic responses to a therapeutic intervention and may serve as potential drug and prognostic targets. Biosensing methods have been a conventional means for biomarker analysis by measuring the binding affinity and rate constants of immunological reactions.

Amerigo Scientific offers a new biomarker analysis service that utilizes the unique patented fiber optic particle plasmon resonance (FOPPR™) technology for real-time antibody-antigen interaction analysis and data processing, providing kinetic curves of antibody-antigen pairs and affinity analysis. This technology does not require additional antibody adsorption-desorption assays, reducing the complexity of the experimental process and detection cost.

FOPPR™ sensing technique mainly utilizes the multiple total internal reflection (TIR) schemes and the evanescent wave to enhance the absorption by gold nanoparticles (AuNPs), as well as the signal-to-noise ratio. When light propagates in the fiber core via consecutive TIR, the particle plasmon resonances (PPRs) of immobilized AuNPs are excited by the evanescent field at the fiber core surface, thus attenuating the light transmitted through the fiber by interaction with AuNPs. The PPR also known as localized surface plasmon resonance (LSPR) is the collective electron oscillations of metal nanoparticles. When the incident photon frequency is resonant with the oscillation frequency of conductive electrons, the absorption and scattering of the electromagnetic radiation are thus enhanced. Hence, the FOPPR™ sensor for monitoring of molecular interactions is based on the localized evanescent field absorption by the AuNPs upon biomolecular interaction. As PPR is extremely sensitive to the change in the local refractive index occurring at the nanoparticle surface, bioreporter molecules are used in the FOPPR™ system to monitor binding biomolecules on the nanoparticle surface and their subsequent affinity interactions in real-time.
