pAAVdual-TUBA1A-EGFP is used to produce AAV-TUBA1A-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a TUBA1A promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-TUBA1A-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The TUBA1A promoter is a neuronal-specific regulatory element that drives the expression of the Tubulin Alpha-1A gene, essential for microtubule function in neurons. It is a critical tool in neuroscience research, particularly for studying neurodevelopment, neurodegenerative diseases, and the roles of microtubules in neuronal function.
