pAAVdual-NES-EGFP is used to produce AAV-NES-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a NES promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-NES-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The NES promoter consists of the rat Nestin intron 2 enhancer fused to the mouse Hsp68 minimal promoter. It is active in neural progenitor and stem cells, enabling selective AAV-mediated expression during early stages of neural development. This promoter is widely used in neuroscience research for lineage tracing, stem cell targeting, and regenerative strategies in the brain and spinal cord. Its strong activity in proliferative neural populations makes it a valuable tool for developmental studies and neural repair applications. Please contact us at for specific academic pricing.
