pAAVdual-K14-EGFP is used to produce AAV-K14-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a K14 promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-K14-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The K14 promoter is an epithelial-specific regulatory element that drives gene expression in the basal layer of the epidermis and other stratified epithelia. It is a critical tool in skin and epithelial research, gene therapy for skin disorders, and the creation of transgenic models to study skin development and disease.
