pAAVdual-hBEST1-EGFP is used to produce AAV-hBEST1-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a hBEST1 promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-hBEST1-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The hBEST1 promoter refers to the promoter region of the human Bestrophin 1 (BEST1) gene, which is primarily expressed in the retinal pigment epithelium (RPE) of the eye. The BEST1 gene encodes a protein that functions as a chloride channel and plays a crucial role in maintaining the health and function of the RPE, which is essential for normal vision. The hBEST1 promoter is a retinal pigment epithelium-specific regulatory element that drives gene expression in the RPE, a vital layer of the retina. It is a crucial tool in retinal gene therapy, ophthalmology research, and the development of transgenic models for studying the RPE and related retinal diseases. Its specificity to the RPE makes it invaluable for targeting gene expression in studies focused on retinal health, disease, and therapy. Please contact us at for specific academic pricing.
