pAAVdual-ApoE/hAAT-EGFP is used to produce AAV-ApoE/hAAT-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a ApoE/hAAT promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-ApoE/hAAT-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The ApoE/hAAT promoter is a chimeric promoter combining elements from the Apolipoprotein E (ApoE) promoter and the human Alpha-1 Antitrypsin (hAAT) promoter. This hybrid promoter is designed to drive gene expression in a liver-specific manner, capitalizing on the strengths of both parent promoters. The ApoE/hAAT promoter takes advantage of the liver specificity and strong expression capabilities of both the ApoE and hAAT promoters. It is often used in gene therapy vectors to ensure that the transgene is expressed predominantly in the liver.
