pAAVdual-4XGRM6-EGFP is used to produce AAV-4XGRM6-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a 4XGRM6 promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-4XGRM6-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The 4XGRM6 promoter is a synthetic, enhanced version of the GRM6 promoter, featuring four tandem repeats to drive stronger and more specific gene expression in ON bipolar cells of the retina. It is a powerful tool in retinal gene therapy, vision research, and the development of transgenic models for studying visual signal processing and retinal diseases.
