pAAVone-AAV-PHP.eB-CMV-EGFP-2.2kb is a plasmid used to produce AAV-PHP.eB-CMV-EGFP using novel AAVone production system. It integrates all the AAV production necessary components, including Ad helper genes (E2A, E4orf6 and VA RNA), AAV helper genes (Rep and AAV-PHP.eB), and a 2.2kb single-strand AAV-CMV-EGFP genome. AAV-PHP.eB-CMV-EGFP viruses can be simply and efficiently produced by transfection of this single-plasmid into host cells, such us HEK 293 and HEK 293T. AAV-PHP.eB is an engineered variant of the AAV9 serotype that was developed to improve the transduction efficiency of the central nervous system (CNS), particularly in mice. It was designed through directed evolution to cross the blood-brain barrier (BBB) with significantly enhanced efficiency compared to the natural AAV9 serotype, making it a valuable tool for gene delivery to the brain and spinal cord.
