pAAVdual-MP-84-EGFP is used to produce AAV-MP-84-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a MP-84 promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-MP-84-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The MP-84 promoter is a micro-promoter derived from the promoter of human insulin gene. Despite its compact size (84 base pairs), it demonstrates robust activity that is comparable to, or only slightly weaker than, the much larger CAG promoter across a wide range of cell types and tissues both in vitro and in vivo. Strong expression has been documented in human islet endocrine cells, hepatocytes, brain, and muscle tissues, among others. By conserving AAV packaging capacity, micro-promoters like MP-84 enable the delivery of larger therapeutic genes that cannot be accommodated with conventional large universal promoters.Please contact us at for specific academic pricing.
