pAAVdual-MP-135-EGFP is used to produce AAV-MP-135-EGFP virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a MP-135 promoter and an EGFP reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-MP-135-EGFP viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The MP-135 is a micro promoter derived from the promoter of human glucagon gene. Despite its small size (135 bp), it demonstrates activity comparable to the strong, ubiquitous CAG promoter across diverse cell types and tissues. MP-135 drives robust expression in both skeletal and cardiac muscle, making it particularly valuable for neuromuscular disease models and cardiac gene therapy. rAAV constructs containing MP-135 have also shown strong activity in cultured cells from all three germ-layer lineages, with reporter expression documented in human primary hepatocytes, pancreatic islets, and multiple mouse tissues in vivo, including brain and skeletal muscle.
