pAAVdual-CaMKIIα(0.36)-Cluc is used to produce AAV-CaMKIIα(0.36)-Cluc virus using novel AAVdual production system. In this plasmid, the regular single strand AAV genome with a CaMKIIα(0.36) promoter and an Cluc reporter is cloned into our novel Ad helper plasmid, mini-pHelper. AAV-CaMKIIα(0.36)-Cluc viruses can be generated by co-transfection of this plasmid with regular AAV helper (pRCap) plasmids, carrying AAV2 rep gene and different cap genes, without adding additional Ad helper plasmid to supply E2A, E4orf6 and VA RNA functions.
The 0.36kb CaMKIIα promoter refers to a shortened version of the promoter region for the Calcium/Calmodulin-Dependent Protein Kinase II Alpha (CaMKIIα) gene. CaMKIIα is a crucial enzyme in the brain, particularly in neurons, where it plays a significant role in synaptic plasticity, learning, and memory. This 0.36kb version of the promoter is specifically designed to drive gene expression in neurons, particularly in excitatory neurons of the forebrain, such as those in the hippocampus and cortex.
Cypridina luciferase (Cluc), a bioluminescent reporter protein derived from the marine ostracod Cypridina noctiluca. Cluc emits light in the blue range when it catalyzes the oxidation of its substrate, Cypridina luciferin. This property makes Cluc an excellent tool for various biological research applications, particularly in non-invasive imaging and real-time monitoring of biological processes. AAV-Cluc is a powerful tool for researchers looking to perform sensitive, non-invasive imaging and real-time monitoring of gene expression and biological processes. Its high sensitivity and low background make it particularly useful for in vivo studies and applications where continuous, non-destructive observation is required. Please contact us at for specific academic pricing.
