Necrostatin-1 is a potent inhibitor of necroptosis, a programmed and caspase-independent cell death mechanism. The death domain receptor-associated serine/threonine kinase RIP1 is essential for TNF-induced necrosis. Necrostatin-1 inhibits RIP1 and blocks TNFα-induced necrosis of FADD-deficient Jurkat cells with an EC50 value of 490 nM.
