MitoTracker® Red CM-H2XRos is a cell-permeable dihydro-X-rosamine derivative containing a weakly thiol-reactive chloromethyl functional group for labeling mitochondria. It is a reduced red fluorescent dye (Ex=579 nm, Em=599 nm) that does not fluoresce. It is passively transported across the cell membrane by simple incubation with cells, where it is oxidized to a fluorescent, mitochondria-selective probe that accumulates on active mitochondria. Once stained, the mitochondria can be fixed (with aldehyde fixatives such as formaldehyde) and permeabilized (with aldehyde detergents such as Triton X-100) as required for subsequent experiments, while the probe remains intracellular. It is suitable for dual-labeling experiments because its red fluorescence has good resolution with other green fluorescent probes.
Although traditional mitochondrial fluorescent probes such as TMR and rhodamine 123 can also easily accumulate on functional mitochondria, they are washed away once the mitochondrial membrane potential is lost, which greatly limits their application in some experiments that require cells to be fixed with aldehydes or contain factors that affect mitochondrial energy status.Please contact us at for specific academic pricing.
