MitoTracker® Deep Red FM is a cell-permeable, carbocyanine-based, deep infrared red fluorescent probe (Ex=644 nm, Em=665 nm) containing a weakly thiol-reactive chloromethyl functional group that labels mitochondria. It can be passively transported across the cell membrane and directly aggregated on active mitochondria by simply incubating the cells. Once the mitochondria are stained, they can be fixed (aldehyde fixatives such as formaldehyde) and permeabilized (aldehyde detergents such as Triton X-100) according to the needs of subsequent experiments, and the probe remains in the cell. This product is suitable for dual-labeling experiments because its red fluorescence has good resolution with other green fluorescent probes. Although traditional mitochondrial fluorescent probes such as TMR and rhodamine 123 can also easily aggregate on functional mitochondria, they will be washed away once the mitochondrial membrane potential is lost, which greatly limits their application in some experiments that require cells to be fixed with aldehydes or contain factors affecting mitochondrial energy status.
