LysoSensor® series probes are a type of fluorescent probes that detect the pH value of acidic compartments (such as lysosomes, acrosomes, and spermatozoa) in living cells. The staining of this type of probe is acidophilic and can accumulate in acidic organelles through protonation. This acidophilicity of this type of probe can relieve the fluorescence quenching of the dye, making the fluorescence intensity emitted stronger. Therefore, compared with the LysoTracker series probes, the fluorescence intensity of the LysoSensor® series probes increases in a pH-dependent (acidophilic) manner as the organelles become acidic.
This product, LysoSensor™ Yellow/Blue DND-160 (PDMPO), is a ratiometric probe for determining the pH value of lysosomes. It is also a pH-dependent fluorescent probe in the LysoTracker® series of probes that displays dual excitation and dual emission wavelengths. The probe produces blue fluorescence (Ex/Em=329 nm/440 nm) in a neutral environment, but emits yellow fluorescence (Ex/Em=384 nm/540 nm) when the environment becomes slightly acidic.
