        • Product Principle
FFPE Sample Release Reagent provide a simple, safe and effective method to release and preserve nucleic acid from FFPE samples. This reagent releases total nucleic acids (DNA and RNA) from cells by lysing the cells with surfactants in an alkaline environment. This reagent can quickly release nucleic acid from FFPE samples.

          Product features:
          · Fast speed.
10 minutes for lysis and release of nucleic acids from paraffin-embedded tissue samples.
          · High safety.
The product does not contain xylene and other dewaxing components, effectively protecting the safety of the experimental operators.
          · Easy to operate.
No need to dewax the paraffin-embedded tissue samples with xylene and nucleic acid extraction and purification, the operator can directly extract the mixture of FFPE samples and this product and add it directly to the PCR reaction system as amplification template for subsequent detection.
          · Long storage time for DNA.
Under room temperature, the DNA released after lysis of paraffin-embedded tissue samples can be stored stably in this product for 3~4 days.

          Main component
It is mainly composed of Tris-HCl(PH8.0)、0.5M EDTA(PH8.0)、Triton X-100 and H2O

          Method of Application
          For Sections of FFPE samples:
          · Scrape off the paraffin-embedded tissue on the slide with a sterilized instrument, submerge the sections into this product (at least three 5 µm thick sections should be added).
          · Tighten the cap of the tube and heat it at 90°C for 10 minutes.
          · Place the product on an oscillator and shake vigorously for a few seconds, then centrifuge to make sure there is no residual material on the wall of the tube.

          Note
          · Please read the manual carefully before using this product. This product is suitable for lysis and nucleic acid preservation of paraffin-embedded tissue samples, and does not have diagnostic and therapeutic functions; check whether the package is damaged before use, If it is damaged, it is strictly prohibited to use.
          · When scraping paraffin-embedded tissue samples from different sources, the blades need to be changed to avoid cross-contamination.
          · If the paraffin content of the sample to be tested is high, flocculent material will be suspended or settled at the bottom of the tube after centrifugation, and the flocculent material should be avoided when taking the sample to avoid blocking the pipette tip and causing inaccurate sampling and other results that may affect the subsequent assay.
          · When performing PCR and other assays on paraffin-embedded tissue samples treated with this product, it is recommended to choose to increase the loading volume (replacing the H2O in the PCR reaction system with the same volume of the mixed solution treated with this product) if the sensitivity of the assay is to be improved.
          · FFPE samples will have a low probability of deamidation of cytosine (C) to uracil (U) in their base sequences during the preparation process, which will lead to the conversion of C>T or G>A in the subsequent PCR reactions, so it is recommended to add UNG enzyme to the PCR system to avoid such phenomenon.
          · This product does not have the function of long-term preservation of paraffin-embedded tissue RNA. If the detection target is RNA, the sample solution treated by this product should be used within 1 hour.
          · Liquid in this product is alkaline, the liquid in the preservation tube should be avoided to splash into the eyes when using, and should be rinsed with plenty of water immediately if it accidentally gets into the eyes.
          · The use of out-of-date products is strictly prohibited; after use, the product should strictly comply with the requirements of relevant laws and regulations and be disposed of in accordance with the classification of medical waste, do not throw away or mix into domestic waste.
          · Repeated freezing and thawing will seriously affect the cracking and preservation effect of the product; it is recommended that the number of freezing and thawing after putting in the sample should not exceed 2 times.
          · Improper storage of samples after collection will result in serious contamination, which will affect the final test results.
          · Failure to store the collected samples at the specified temperature will affect the final test results.
          · This product is suitable for the use of trained experimenter. Gloves and masks should be worn for protection.
          · The used product should be disposed of according to local, state, and federal regulations. All wastes containing the material should be properly labeled.
          · The test results of the samples are related to the quality of the samples collected, handled, and shipped. Any error in this process may result in a false negative result.
          · Ensure biosafety requirements are met in accordance with the local regulations throughout the entire procedure.

      • Applications
        • Application
          For PCR, qPCR, etc.
          Application Description
This product is used for the pretreatment of FFPE samples (paraffin-embedded tissue samples), so that the nucleic acid (DNA&RNA) to be tested can be released from the binding state with other substances. In this way, nucleic acid can be easily and directly detected. The crude lysates can be directly applied in polymerase chain reaction (PCR) , RT-PCR and digital PCR, etc.
