B4GALT1 (Y289L) is a recombinant, bovine-derived mutant galactosyltransferase. It is used for enzymatic coupling of specific N-glycans to O-GlcNAcylated proteins or for transferring azide-containing N-acetylgalactosamine (UDP-GalNAz) to exposed GlcNAc residues in protein engineering workflows.
Workflow:
· Deglycosylation:
rEndo S2 glycosidase removes N-glycans from antibodies, leaving the Fc glycan with exposed GlcNAc residues.
· Azide Activation:
B4GALT1 (Y289L) transfers UDP-GalNAz to the exposed GlcNAc residues, generating azide-activated antibodies that can react with alkyne-functionalized tags.
· Click Chemistry Conjugation:
Azide-activated antibodies undergo copper-free click chemistry (SPAAC) with cyclooctyne-functionalized labels, forming stable triazole linkages at GlcNAz sites. Further azide-labeled antibodies can be conjugated with fluorescent tags or other functional moieties via SPAAC click chemistry. Please contact us at for specific academic pricing.
