Background
Ac-DEVD-pNA is a colorimetric substrate for caspase-3 (Km=9.7µM) and caspases-1, -4, -6, and -7 [1]. Ac is the acetyl group and pNA is p-nitroanilide. Sequence is based on PARP (poly(ADPribose) polymerase) cleavage site Asp216 for caspase-3. In apoptosis, activated caspase-3 cleaves substrates such as poly(ADP-ribose) polymerase that targets at the amino acid sequence DEVD [2]. Cleavage is monitored colorimetrically at 405nm. Ac-DEVD-pNA can be used to identify and quantify caspase-3 activity in apoptotic cell lysates.
