TMB is commonly used as a chromogenic substrate for horse radish peroxidase for detection purposes in assay formats, such as enzyme-linked immunosorbent assays (ELISAs). TMB is a colourless solution that changes to blue upon oxidation. In the ELISA format, a stopping solution (such as sulfuric acid) is typically added to a TMB-containing solution to prevent extensive colour generation. After addition of the stop solution, the TMB solution changes from blue through to yellow (450 nm), generating a product that yields a high molar absorptivity. TMB chromogenic substrate is selected for its high sensitivity and stability.
