T4 Phage β-glucosyltransferase can transfer the glucose group of uridine diphosphate glucose (UDP-glucose) to 5-hydroxymethylcytosine (5-hmC) residues in double-stranded DNA to generate β-glucosyl-5-hydroxymethylcytosine (5gmC), thereby protecting 5-hydroxymethylcytosine (5-hmC) from cleavage by MunⅠ restriction endonuclease.
