Klenow fragment is a protein hydrolysis product of E. coli DNA polymerase I. It retains the DNA polymerase activity and 3'→5' exonuclease activity of DNA polymerase I, but lacks 5'→3' exonuclease activity. It can be used for random primer DNA labeling and cDNA second chain synthesis.
Unit Definition:
One activity unit (U) is defined as the amount of enzyme required to incorporate 10 nmol of dNTP into acid-insoluble precipitate within 30 minutes at 37°C.
