        • Propidium Iodide (PI) is a fluorescent nucleic acid dye that can be used for staining mammalian cells, bacteria, and yeast. It cannot penetrate the membrane of live cells but can pass through the membranes of dead cells and cells in the late stages of apoptosis to stain the nucleus. Therefore, PI is commonly used as a fluorescent probe for detecting apoptosis or necrosis. Once inside the cell, PI binds to DNA and RNA, increasing fluorescence intensity by 20 to 30 times. In flow cytometry, PI is often used in combination with other dyes such as Calcein-AM, Hoechst 33258, or Hoechst 33342 to distinguish between early and late apoptotic cells as well as dead cells. Additionally, PI is also frequently used as a counterstain in multicolor fluorescence staining.
