pAAVone-AAV4-CMV-mCherry is a plasmid used to produce AAV4-CMV-mCherry using novel AAVone production system. It integrates all the AAV production necessary components, including Ad helper genes (E2A, E4orf6 and VA RNA), AAV helper genes (Rep and AAV4 Cap), and a single-strand AAV-CMV-mCherry genome. AAV4-CMV-mCherry viruses can be simply and efficiently produced by transfection of this single-into host cells, such us HEK 293 and HEK 293T. AAV4 is a lesser-known serotype of adeno-associated virus with a distinct tissue tropism compared to other AAV serotypes. It has a specific affinity for the airway epithelium and certain regions of the brain, such as the ependymal cells that line the brain's ventricular system. This makes AAV4 less commonly used in traditional gene therapy applications, but it has potential for targeting specific tissues where its tropism is beneficial.
