Globin mRNA accounts for about 30%-70% of the mRNA from blood samples, accounting for a large amount of RNA sequencing data. Therefore, the removal of globin mRNA during the construction of blood sample RNA libraries is an effective method to increase the information content of sequencing data. Hieff NGS® One-Step Globin mRNA Removal Kit uses targeted technology to highly specifically inhibit the reverse transcription of globin mRNA in total RNA during cDNA synthesis, without affecting the cDNA synthesis of other mRNAs, non-coding RNAs, and non-target RNAs such as circular RNA, thereby highly specifically removing globin mRNA during the synthesis of one-strand cDNA. This product, combined with Hieff NGS® One-Step rRNA Removal Ki, can quickly remove rRNA and globin mRNA from RNA derived from human blood samples. Compared with the traditional RNase H digestion method, this targeted removal technology has the advantages of short time (4 min), simple operation (one-step operation), and intact non-target RNA. It greatly simplifies the operation process and improves the efficiency of library construction. It can be used in conjunction with various one-strand cDNA synthesis reagents on the market. This kit has a good globin mRNA removal effect for both intact and partially degraded total RNA. The cDNA samples obtained by globin mRNA removal can be used for high-throughput sequencing analysis of mRNA and non-coding RNA, which can significantly increase the proportion of valid data in the sequencing results, and can also be used for other cDNA downstream applications.
