The AAV-sgRNA Selection Kits are designed for efficient sgRNA selection and AAV vector creation. Each kit includes 4 pre-designed sgRNA constructs targeting your gene of interest (GOI), 1 control sgRNA, and a GOI-mCherry fusion reporter plasmid. All plasmids contain two AAV ITRs, enabling the constructs to be directly used for AAV vector packaging after selection. The Cas protein and sgRNA are combined in a single plasmid, and users need to select a Crispr-Cas system to be included in the kit.
Kit Components:
· 4x Custom sgRNAs (targeting the gene of interest, GOI) and 1x Control sgRNA
Expressed under the U6 promoter (strong Pol III promoter for sgRNA expression).
· Single Plasmid System (expressing both sgRNA and Cas protein):
Cas protein under a small but strong ubiquitous promoter.
· GOI-mCherry Fusion Reporter Plasmid:
Allows real-time monitoring of gene knockdown efficiency via mCherry fluorescence.
Workflow for Customers:
· Provide GOI Information:
Submit the target gene name or sequence for custom sgRNA design.
· Select Gene Editing System:
Choose between Cas9, Cas12, Cas13 or other options.
· Co-transfect Plasmids:
Deliver both the AAV-sgRNA/Cas plasmid and the GOI-mCherry reporter into cells.
· Monitor Knockdown Efficiency:
Track mCherry fluorescence (reduction indicates successful knockdown).
Compare the 4 sgRNAs to identify the most effective one.
