Silica reversed phase chromatography (RPC) is a popular purification tool for peptide-based therapeutics such as insulins and GLP-1 compounds. In many cases, these chromatography process need to be performed at high pH conditions, such as column regeneration with 0.1M NaOH solution, where conventional silica media has a short lifetime.
Hybrid silica particles have high pH stability but they are less than 10 µm in size and are only used in analytical HPLC applications. The large size (more than 10µm) of hybrid silica particles for process-scale chromatography is limited by small surface area, high manufacturing cost, and difficulties in large-scale production. Other hybrid coated silica particles have improved lifetime over silica particles at high pH, but still are not as stable as hybrid particles.
Based on the innovative Precisely Controlled Silica (PCS) technology, monodispersed hybrid silica - UniHybrid® Eterne particles in 10 µm size are developed.
