        • Oligodendrocytes in the central nervous systemform myelin sheaths around neuronal axons, which canspeed up the conduction of nerve impulses and providenutritional support to neurons. Abnormal myelination is associated with a variety of central nervous system diseases. With this TrueGold Myelin Staining Kit, the myelin sheath can be stained reddish-brown. Usually, myelin can be stained by immune-histochemical staining of myelin-associated proteins, such as MBP, PLP, MAG, etc., or by dyes such as Luxol Fast Blue. Compared with these methods, the TrueGold Staining is much more faster and simpler, and have better resolution and contrast.

          Manual:
          1. Frozen section at 14~25 μm, and bake your slides at 37~50°C for 15~30 min.
          2. Dilute desired doses of 1x Staining and 1x Stop Solutions with deionized water.
          3. Incubate your sections in 1x staining solution in a humid chamber or other sealed container at 45°C for 20-30 min in the dark. Microscopic monitoring is recommended after 20 min. (Staining time should be adjusted based on section thickness and tissue age).
          4. Rinse the slides twice with deionized water for about 1 min each.
          5. Add 1x Stop solution and incubate at 45°C for 2-3 min.
          6. Rinse the slides 3 times with deionized water for about 1 min each.
          7. Mount your sections and collect data.
          Note: PBS and any other reagents are not required during staining.

          Notice: This kit is used for the staining of frozen sections. It is not suitable for paraffin sections.
