rIdeZ Protease, 50u, Mass Spectra Grade

rIdeS | rIdeZ (Immunoglobulin G-specific Proteases) are engineered recombinant proteases overexpressed in E. coli. They specifically cleave IgG molecules at a single site below the hinge region, producing homogeneous F(ab´)₂ and Fc fragments. Both IdeS and IdeZ efficiently cleave human IgG1, IgG2, IgG3, and IgG4, as well as IgG from monkey, sheep, and rabbit, and humanized or chimeric IgG, along with Fc-fusion proteins. However, only IdeZ is capable of cleaving mouse IgG2a and IgG3.

Physical Form: Lyophilized powder.
Reconstitution: Dissolve in 40 µL of ultrapure water before use.
Shelf Life: 24 months when stored at –20°C.
pH Range: Optimal activity between pH 6–8.
Application: rIdeS | rIdeZ proteases are IgG-specific proteases that generate homogeneous F(ab´)₂ and Fc fragments.

Digestion SOP:
· Prepare 10 µL of mAb sample (10 µg/µL).
· Add 5 µL of 10× Digestion Buffer (HLS PBS001) and 31 µL of ultrapure water, for a total reaction volume of 50 µL.
· Add 4 µL of rIdeS protease (0.5 µg/µL).
· Incubate at 37°C for 30 minutes.
