Figure 1. Agonist-induced Serine425/Serine429 phosphorylation of VPAC2 Receptor. Upper panel, HEK293 cells stably expressing the VPAC2-Receptor (VPAC2) were either not exposed or exposed to 1 µM of specific VPAC-Receptor agonist PACAP-27 for 30 minutes. Cells were lysed and immunoblotted with the anti-pS425/pS429-VPAC2 antibody (7TM0372C) at a dilution of 1:1000. Lower panel, blot was stripped and reprobed with the phosphorylation-independent anti-VPAC2 antibody (7TM0372N) at a dilution of 1:1000 to confirm equal loading of the gel.

Note, Serine408/Serine409-phosphorylated VPAC2 receptors were not detectable in untreated cells (0 min). Serine425/Serine429-phosphorylated VPAC2 receptors were seen at the plasma membrane after 5 min PACAP-27 stimulation.
