NHS-Activated MagBeads are pre-activated polymer magnetic microspheres with NHS groups modified on the surface. They can be coupled to amino-containing proteins or peptides. They are mainly used for affinity purification or analytical detection of antibodies, antigens and other biomolecules. It provides a convenient way to couple any desired protein to the surface of magnetic beads. Compared with traditional carboxyl and amino magnetic beads, magnetic beads with NHS groups on the surface do not need to be activated with EDC/NHS or glutaraldehyde in advance. Simply dissolve the amino-containing biological ligand in the coupling buffer and incubate the protein solution with the magnetic beads for 2-4 hours to covalently couple the biological ligand to the magnetic beads through NHS-ester chemistry. This process does not require hazardous chemicals or lengthy reaction schemes, and has the advantages of simple operation, mild coupling conditions, and fast and efficient biological ligand coupling.
The activated magnetic beads contain N-hydroxy-succinimide (NHS) functional groups that can react with primary amines to form stable amide bonds. Once they are covalently linked, the fixed proteins will not leach from the surface of the magnetic beads. When using prepared magnetic beads in experiments, non-specific binding is negligible because the non-reactive NHS-ester groups are completely blocked during the coupling procedure. NHS-activated magnetic beads can be coupled and handled using a manual magnetic stand.
